A non-invasive, outpatient device that temporarily opens the blood-brain barrier (BBB) to enable therapeutic delivery of drugs, biologics, and gene therapies into the brain — without requiring redesign of the molecule. The 4-step outpatient procedure (45-minute non-invasive BBB priming, standard IV infusion, enhanced delivery, and same-day discharge) requires no sedation, shaving, or surgery. The platform has demonstrated delivery of payloads ranging from small molecules (55 kDa) to antibodies (150 kDa) and viral vectors (5,300 kDa) — all above the natural BBB cutoff of 0.4 kDa that blocks 98% of CNS drugs. Validated across rodent, porcine, and non-human primate models, with NHP data showing 6x (up to ~800% of wild-type) ARSA enzyme activity in the substantia nigra following AAV delivery.
